Cytogenetic Analyses of Leontopithecus chrysopygus (Cebidae, Platyrrhini)

Resumo

The genus Leontopithecus (Callitrichinae, Cebidae, Platyrrhini) reunites four species, all classified as endangered by the IUCN. The black lion tamarin Leontopithecus chrysopygus is endemic to the Atlantic Forest and currently found in the state of São Paulo. In order to compare the karyotype of L. chrysopygus (2n=46, FN=74) with those of the other species of the genus, we analyzed the chromosomes of a male and a female L. chrysopygus after GTG- and CBG-banding, silver staining of the nucleolus organizer regions (AgNOR) and fluorescence in situ hybridization (FISH) with telomeric, 18S rDNA, and alpha satellite DNA probes. The results were compared to those available for L. rosalia and L. chrysomelas. The three species presented great similarity in their GTG-banding patterns and in the centromeric localization of the alpha satellite DNA. They mainly differed in their constitutive heterochromatic and NORs distribution. The genomes of L. chrysopygus and L. rosalia were compared through interspecific FISH using the total genomic DNAs of each species as probes (GISH). The results showed extreme conservation among the euchromatic part of the genomes and differences in the heterochromatic portions containing species-specific sequences in Leontopithecus. Further analysis of these sequences may contribute to the identification of species-specific markers.
